N-糖苷酶 F (PNGase F)酶活定义(UnitDefinition)1个酶活力单位指在10μL的反应体系中,37℃条件下1小时从10μg变性RNaseB中除去超过95%的碳水化合物所需要的酶量。储存条件-15~-25℃保存,有效期1年。使用说明变性条件下蛋白质去糖基化1)在水中加入1μLBuffer1和目标糖蛋白(1-20μg),至终体积10μL;2)100℃温度下煮沸10min使其变性,冰上冷却,离心10秒;3)加入2μL的Buffer2、2μL的10%NP-40、6μL去离子水,总反应体积20μL;4)加入0.2~0.5μL的PNGase,轻轻混匀。在37℃孵育1-3h。非变性条件下蛋白质去糖基化1)在水中加入2μL的Buffer2和目标糖蛋白(1-20μg)至体积为20μL。2)加入0.5~1μL的PNGaseF,轻轻混匀。3)37°C孵育4-24h。注意:在变性条件下大多数底物能够更好的去糖基化,在非变性条件下可能需要增加PNGaseF的量和延长孵育时间。Recombinant Human LILRB4/CD85k/ILT3(hFc Tag)
